What is Prezcobix (Darunavir/Cobicistat)?

Prezcobix (brand name) is a medication containing two drugs, darunavir and cobicistat. Darunavir is a human immunodeficiency virus (HIV-1) protease inhibitor and cobicistat is a CYP3A inhibitor. The generic name is darunavir and cobicistat. It is indicated for the treatment of HIV infection. It is used with other antiretroviral medicines. The manufacturer of this product is Janssen.

What is this medication used for?

Prezcobix is used to treat both treatment naïve and treatment experienced patients with no darunavir resistance-associated substitutions. The efficacy and safety of Prezcobix has not been established in pediatric patients, so this drug should not be administered to children. This drug was also not adequately assessed in geriatric patients (≥ 65 years of age) so older patients considering treatment with this medication should consult closely with a physician to determine if this course of treatment is right for them. Elderly patients are at greater risk of decreased hepatic function with this type of treatment.

How does this drug work?

The two-drug combination of this product is darunavirand and cobicistat. Darunavir works by inhibiting the HIV-1 protease by selectively inhibiting the cleavage of polyproteins in infected cells. As a result, mature virus particles are prevented from forming. Cobicistat is an inhibitor of cytochromes P450 of the CYP3A subfamily. It is a mechanism-based inhibitor.

How to take it

Prezcobix has a usual dose of one tablet that is taken once per day with food. Testing should occur prior to starting treatment with experienced patients. HIV genotypic testing is recommended, and creatinine clearance should be assessed.

Patients should follow a regular dosing schedule to prevent the development of resistance. Therapy should not be altered or discontinued without consultation with a physician.

Prezcobix side effects

The drug may cause liver problems as well as severe or life-threatening skin-reactions or rash. When taken with certain other medications, new kidney problems, worsening of kidney problems, or kidney failure may occur.

Prezcobix is taken as a film coated tablet, once daily, orally with food. This is the recommended dosage. This product contains two active ingredients. It is a fixed-dose combination product containing 800mg of darunavir and 150mg of cobicistat. Prezcobix is administered in conjunction with other antiretroviral agents.
